PRA-prcd (progressive rod-cone degeneration)

The progressive rod-cone degeneration (PRA-prcd) is caused by mutation c.5G>A in PRCD gene (former designated as 1298G>A). The PRCD gene affects mainly retina with the same expression level in retinal pigment epithelium, photoreceptor cells and in the ganglion cells. The mutation causes exchange of nucleon TGC basis for TAC basis that results in binding of other amino acid and structural and functional changes in the resulting protein.

This genetic disorder causes that photosensitive cells of the retina (rods and cones) gradually deteriorate and die. The onset of the disease is in adolescence or in early adulthood. However, the age of the disease onset and the disease intensity depends on the specific breed. The rod-type cells lose their function first and the affected dogs will develop night blindness. Over time the disease continues with cone degeneration and progresses to complete blindness.

The genetic testing can help to clarify the diagnosis. It can determine the dog genotype and is the solution for gradual elimination of prcd occurrence in a breed. It is necessary to be aware that not all retinal diseases belong in progressive retina atrophy group of disorders and not all are a variant of PRA-prcd.  In many breeds, the cause of PRA inheritance has not been still clarified. It is also possible that several mutations can be responsible for retinal atrophy in one breed. Therefore, we recommend that an eye examination by a veterinary ophthalmologist is performed every year.
